Dr. Lu has long been committed to identify the single-molecule defects that lead to disease. The complexity of multiple effects of disease relies on many different types of networks, from protein-protein interactions at the cellular molecular level to gene expression in the organism. We can took cardiovascular calcification in kidney disease as an example, cardiovascular disease is still the main cause of death in end-stage renal patients. The main cause of cardiovascular disease in chronic kidney disease is arterial calcification. In patients with end-stage renal failure, there are many factors that cause vascular calcification. The risk factors include hyperphosphatemia, hypercalcemia, hyperparathyroidism, overdose of active vitamin D3, inflammation, etc. The key mechanisms of vascular calcification are (1) failure of the anti-calcification process due to loss of inhibitors and lack of constitutively expressed mineralization inhibitors leading to vascular calcification. (2) Various stressors induce osteogenic transdifferentiation of VSMCs. VSMCs are the product of matrix vesicles and are the basis for calcium phosphate deposition. (3) Cell death due to apoptosis or necrosis results in the release of apoptotic bodies or necrotic debris, which may act as a nucleation agent for apatite. (4) Abnormal mineral homeostasis leads to the deposition of calcium phosphate hydroxyapatite. (5) The nuclear complex formed during bone remodeling promotes ectopic mineralization. (6) Matrix degradation/modification caused by environmental stress factors is related to vascular calcification. In the past few years, a large amount of investment and clinical trials have been carried out on intermediates of bone and mineral metabolism, and new treatments for calcified substances are actively targeted, hoping that the research results can slow down vascular calcification.
